With Virtual TI or TiLP, but only if you have a non-direct USB cable.

If you have a direct USB cable (USB on both ends), you'll have to reconstruct one using an 8XU file from TI's site (education.ti.com) and the boot code on your calculator, which you can dump (and combine with the 8XU to create a ROM image) using rom8x which is available for download at ticalc.org.

That's that ZAC(for the 86), the OTBP assembler, and now PHasm are for Smile that way you don't even need to transfer the program because its compiled on calc. Though that's really only useful if you are will to lose almost everything in a crash. Backups FTW.
